Marlon Wayans made an appearance along with his siblings on In Living Color between 1992 and 1993. Later on from 1995 to 1999, Wayans co-starred in the WB sitcom The Wayans Bros. alongside his brother Shawn Wayans. Wayans was formerly considered for the role of Robin in the 1992 film Batman Returns. However, the producers cut off the character as there were already too many characters in the movie.

Marlon Wayans was then originally confirmed for the role in the 1995 sequel Batman Forever. He was to play opposite Billy Dee Williams as Two-Face. But the directors changed mid-production from Tim Burton to Joel Schumacher. This led to both parts being recast and Wayans being paid out. He was not signed but his contract for Batman allowed him the option of reprise. He still receives some royalty payments to this day for this. Later, he was substituted by Chris O’Donnell.

It has been stated that he produced the first two films of the Scary Movie series. In this, he and Shawn were credited as writers and co-stars. The films were released in 2000 and 2001. Wayans produced the Nickelodeon cartoon series Thugaboo. In 2017 he was given his own sitcom by NBC, called, Marlon. The show went on for a 10-episode run. Marlon was renewed for a second season by NBC in September 2017. The show is now available on Netflix.
