Folake was born in Victoria Island in Lagos to Nigerian politician Babatunji Olowofoyeku, her father and mother, Felicia Olowofoyeku being the youngest amongst the 20 children. Folake went to Igbinedion Education Center, a Montessori boarding school in Benin City for her primary studies. Later she got admitted to Vivian Fowler Memorial College for Girls in Ikeja and further attended Oxbridge Tutorial College. While studying economics in expectation to become an attorney, Folake received a B.A with honors in theater from City College, New York. While pursuing her BA at City College, she played NCAA Division III college basketball for the CCNY Beavers. In 2001, Folake emigrated to the United States to live with her sister.

After graduating from college, Folake began her career by working at an Off-Broadway theater in New York City. Folake made her guest appearance in roles of television series such as 30 Rock in 2010, How to Get Away With Murder in 2016, Law and Order; Criminal Intent in 2010, and many more. In 2017, Folake played role in the last season of the TV series Transparent, opposite Gaby Hoffmann as her love interest, Lyfe.
