Marpet was not heavily recruited by larger collegiate programs after graduating from high school. By eating 7,000–9,000 calories per day, he boosted his weight from 250 pounds (110 kg) as a freshman to 280 pounds (130 kg) by sophomore year and 290 pounds (130 kg) by junior year. In college, he intended to play both basketball and football.

He was a three-year starter at left tackle for the Hobart football team during his four years on the club. Marpet started 37 of the Statesmen’s 43 games, helping them capture four Liberty League titles in a row. Hobart finished his career with a 41–5 record, only losing one regular-season game.
He was named to Lindy’s preseason All-American first team, a Beyond Sports Network (BSN) preseason All-American, and D3football.com’s preseason All-American second team in 2014. He received the Bill Middleton Memorial Award and the William C. Stiles ’43 Memorial Award. Marpet was a part of the Senior Bowl college football all-star game in 2015.

Marpet’s accomplishments at the NFL Scouting Combine in February 2015 established him as one of the most athletic offensive linemen in the 2015 NFL Draft. Marpet also had the best “Speed, Power, Agility, Reaction, and Quickness” (SPARQ) score out of all the offensive line candidates in the draught. Almost every NFL team was interested in picking Marpet after his outstanding combine performance. On May 1, 2015, the Tampa Bay Buccaneers selected Marpet in the second round, 61st overall, of the 2015 NFL Draft. Marpet started at right guard for the Tampa Bay Buccaneers in his debut season in 2015. He had the highest pass-blocking efficiency of any rookie guard according to PFF. During the 2017 season, the team switched Marpet from guard to center, replacing previous starting center Joe Hawley. Marpet signed a five-year, $55.125 million contract deal with the Buccaneers that runs through the 2023 season, with $27.125 million in guaranteed money, on October 9, 2018. In 2019, he was chosen to the Pro Football Focus All-Underrated Team. During the 2020 regular season, Marpet appeared in 13 games, starting 13 of them. Marpet was a key part of the Buccaneers’ Super Bowl LV-winning postseason run, appearing in all four games.

Marpet announced his retirement from professional football at the age of 28 after seven seasons on February 27, 2022, via an Instagram post. He retired with two years left on his contract and a $10 million annual compensation. Many of Marpet’s former teammates, including Tom Brady, congratulated him on his retirement via social media.
